Product information
-
Function
P-ManNDAz is an unnatural diazirine-containing monosaccharide that acts as a photo-crosslinker. The diazirine group is activated upon UV radiation. 1
-
Mode of action
P-ManNDAz is taken-up by cells and converted to CMP-SiaDAz inside cells. It is incorporated into cell surface glycoproteins and glycolipids. 1
-
Applications
P-ManNDAz is a cell-permeable photo-crosslinking probe. It can be used to investigate the role of glycoproteins, glycolipids and their interactions.
-
Handling
P-ManNDAz is soluble in DMSO and PBS and is added to cell culture from a stock solution. Incubation of the cells with P-ManNDAz for 72 hrs leads to detectable levels of Neu5NDAz residues at the cell surface. Sialoglycans decorated with Neu5DAz residues can be photo-crosslinked using 350 nm light for 20 min to achieve a cross-link between the Neu5NDAz modified protein and its binding partner (for example a Siglec receptor). 1
