Function
Copper Sulfate, Sodium Ascorbate and THPTA can be used to perform the copper-catalyzed azide-alkyne cycloaddition reaction (CuAAC) between an azide and alkyne. Copper Sulfate and Sodium Ascorbate are combined to produce the active Copper(I) catalyst. THPTA is a water soluble ligand that reduces the cytotoxicity of the Copper(I) catalyst.

Mode of action
Copper Sulfate is reacted with Sodium Ascorbate to product the active Copper(I) catalyst. Copper ligand THPTA accelerates the copper-catalyzed azide-alkyne cycloaddition reaction (CuAAC) used to selectively conjugate functional handles to azide or alkyne on live or fixed cells. The copper catalyst reagent is cytotoxic due to the formation of reactive oxygen species (ROS).12 THPTA reduces the time needed for the bioorthogonal labeling step and also allows for a lower concentration of (cytotoxic) copper catalyst. THPTA significantly reduces cytotoxicity the bioorthogonal azide-alkyne click reaction.13
